Comparative Analysis of Phytochemical Contents and Antioxidant Activities of Leaf Extracts from Centella asiatica (L.) Urban. and Hydrocotyle umbellata L.

Abstract

Centella asiatica is a medicinal plant with therapeutic importance, because it can be consumed for neuroprotective effects, anti-hyperglycemia, antioxidant activities, and anti-inflammatory effects. However, consumers mostly confuse C. asiatica with Hydrocotyle umbellata because of their similarity in morphological characters. Therefore, the present study aimed to compare the total phenolic content (TPC), total flavonoid content (TFC), and antioxidant activity using DPPH assay of leaf extracts from C. asiatica and H. umbellata, as well as to evaluate the efficiency of two solvents, which were ethanol and methanol in extracting phytochemical compounds from the leaves of both species. The results revealed that C. asiatica leaf extracts contained a higher TPC than H. umbellata leaf extracts. The extracts with ethanol yielded a greater TPC than methanol. The ethanolic extract of C. asiatica leaves exhibited the highest TPC, at 930.50 mg GAE/g extract, whereas the methanolic extract of C. asiatica leaves showed a total flavonoid content of 1,060.13 mg QE/g extract, which exceeded that of the ethanolic extract. However, the ethanolic and methanolic extracts of H. umbellata leaves yielded comparable TFC. The comparison of solvent efficiency revealed that methanol was more suitable than ethanol for the extraction of flavonoid compounds. Considering antioxidant capacity using DPPH assay, C. asiatica leaves displayed higher antioxidant activity than H. umbellata leaves, while methanolic extraction provided greater extraction efficiency of antioxidants than ethanolic extraction. The methanolic extract of C. asiatica leaves demonstrated the highest antioxidant activity among all groups, with an IC50 value of 15.74 µg/mL. These findings highlight the importance of identification and selection the correct plant species, as well as choosing an appropriate extraction solvent, since these factors may influence the yield of phytochemical compounds and their beneficial biological activities, thereby maximizing the efficacy delivered to consumers.
