Anti-quorum sensing activities of actinobacteria against Serratia marcescens

Bacterial quorum sensing (QS) is a cell-to-cell communication system that regulates a wide range of physiological processes, including biofilm formation and the production of virulence factors. Therefore, quorum sensing inhibition (QSI) has emerged as a promising strategy for controlling bacterial infections. The aim of this study was to investigate the anti-quorum sensing activity of actinobacterial isolates against Serratia marcescens. Among ten actinobacterial isolates screened, only isolate PKS22-27 exhibited QSI activity, as evidenced by inhibition of prodigiosin pigment production, while showing no antibacterial effect on the growth of S. marcescens. Secondary metabolites of PKS22-27 were extracted from cell-free supernatant using ethyl acetate and further evaluated the anti-QS activity by an agar well diffusion assay. The crude extract consistently suppressed pigment production without inhibiting bacterial growth, confirming its quorum sensing inhibitory potential. Identification based on partial 16S rRNA gene sequence analysis revealed that isolate PKS22-27 belonged to the genus Streptomyces and showed the highest sequence similarity to Streptomyces badius JCM4350T (99.14%). These findings indicate that Streptomyces sp. PKS22-27 is a promising source of quorum sensing inhibitory compounds. The inhibition of prodigiosin production, a QS-regulated trait, suggests a potential effect on biofilm formation. Therefore, further biofilm formation assays are required to confirm the underlying mechanism of action.
